|dc.description.abstract||BRAF is a human gene that makes a protein called B-Raf, which is involved in sending signals inside cells related in directing cell growth. In the mutation of B-RafV600E gene, valine, six hundredth amino acid of the BRAF gene is substituted to glutamic acid. Mutation in B-RafV600E gene is regarded as an oncogene and the mutation is especially discovered in 70~80% of papillary thyroid carcinoma with bad prognosis. In this study, human fibroblast was B-RafV600E mutated with the lentivirus. The morphology, growth rate, senescence marker(SAβ-galstaining) of the normal and mutated cell was measured and compared. The mutated cell showed senescent form of large and flat morphological change and the rate of cell growth decreased compared to wildtype cell. Also, the mutated cell was more stained from SAβ-gal staining which shows the senescence of the cell. Same result was verified in the further experiments with human thyrocyte. The result shows the mutation B-RafV600E gene it self makes cell senescent that eventually leads to apoptosis, and the reshould be further stimulations with the mutation to in duce cancer.||-|
