Protective effect of JAK3 inhibition on growth factor deprivation induced neural progenitor cell death

Abstract
Apoptosis is evolutionally conserved, and plays central roles during normal development of the nervous system. The number of embryonic neuroepithelial cells (NECs) is regulated by programmed cell death when NECs are substantially proliferated by growth factors(GFs). The extrinsic factors such as epidermal growth factor (EGF), basic fibroblast growth factor(bFGF), as well as intrinsic protein regulate growth, proliferation and differentiation. Therefore, in culture system, addition of both bFGF and EGF is critical to maintain and expand NECs. The withdrawal of the seGFs induced apoptosis through intrinsic mitochondrial pathway in NECs. However, the mechanisms underlying the fate of NECs to either survive or dieare incompletely understood. In the brain, a specific JAK3 inhibitor aids in the survival of a transgenic amyotrophic lateral sclerosis mouse model. We previously reported that JAK3 inhibition induced neuronal differentiation accompanied by neurite outgrowth in enriched condition of GFs such as EGF, bFGF. In the present study, effect of JAK3 inhibition was examined on GFs withdrawal induced death of cultured NECs from embryonicmice brain (E13.5 days). The GFs withdrawal decreased cell vibility in time-dependent manner, however treatment of JAK3 inhibitor (WHI-p131) increased cellviability. The try pan bluestained cells, which were dead, were increased in GFs-withdrawal condition, incontrast those were decreased by JAK3 inhibitor. For further confirmation, we also performed propidiumiodide (PI) and calcein-AM double staining. The number of calcein-AM stained cells, which were alive, was increased strikingly by JAK3 inhibitor. However, the number of PI-positive cells, which were dead, was similar between JAK3 inhibitor treated and non-treated condition. To confirmidentity of survival cells, the cells were stained with immunocytochemical markers for several cell types. JAK3 inhibitor increased the number of nestin, SOX2 (markers of NPCs) and MAP2(a marker of neuron) positive cells. But capability of proliferation of NSCs was not affected by JAK3 inhibitor. GFs withdrawal is known to induce apoptosis, which could detect by TUNEL(terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nickendlabeling)assay. The number of TUNEL positive cells was decreased in a JAK3 inhibitor dose dependently. And we confirmed casapse-3 activity, since caspase-3 is a caspase protein that plays a central role in the execution of cell apoptosis. Caspase-3 activity that was not affected by JAK3 inhibitor was decreased by caspase-3 inhibitor, but cell viability was not changed by caspase-3 inhibitors. In GFs withdrawal condition, phosphorylation of JAK3 was increased but treatment of JAK3 inhibitor reduced phosphorylation of JAK3. where sGFs withdrawal resulted in decrement of phosphorylation of AKT, ERK and was increment pattern of phosphorylation of JNK, JAK3 inhibitors shown a tendency to increase phosphorylation of AKT and more decreased phosphorylation of ERK, JNK than non treated group. And, JAK3 inhibitor shown a tendency to decrease of Bim-EL protein level, which is BH3-only protein of pro-apoptotic bcl-2 family. And, expression of adhesion molecules such as N-cadherin, NCAM was increased by JAK3 inhibitor. In summary, we identified that GF withdrawal induced apoptosis of NSCs, but inhibition of JAK3 drives NSCs toward cell survival. Also, inhibition of JAK3 promotes NSCs to differentiate into mature neuron accompanied by neurite outgrowth via increase of neuronal adhesion molecule expression such as N-cadherin, NCAM. Therefore, we propose that JAK3 may have acritical role on not only survival but also differentiation of NSCs.

아폽토시스(apoptosis)는 진화론적으로 그 기전이 보전되어 나타나는 세포사멸과정으로 정상적인 신경계 발달과정에서 중요한 역할을 하는 것으로 알려져 있다. 신경발생과정(neurogenesis)에서 줄기세포의 성격을 지닌 신경상피세포(neuroepithelial cell)는 스스로 증식하여 분열을 하는데, 세포 증식을 통해 증가한 신경상피세포의 수는 아폽토시스와 같은 세포예정사(programmed cell death)에 의해 개체 내에서 적절히 조절된다. 또한, 신경상피세포는 스스로 증식하고 신경세포로 분화하고 성장하기 위하여 내부 단백질을 비롯하여 EGF (epidermal growth factor)와 bFGF (basic fibroblast growth factor) 같은 외부 성장인자를 필요로 한다. 따라서 신경상피세포의 실험적인 배양환경에 EGF와 bFGF를 배지에 첨가하여 배양한다. 그리고 이 배양환경으로부터 성장인자의 제거는 내인성 미토콘드리아 경로(intrinsic mitochondrial pathway)를 통한 신경상피세포의 아폽토시스가 유도된다. 이처럼 발생과정에서 나타나는 세포의 죽음 또는 생존과 관련하여 선행 연구자들에 의해 많은 연구가 진행되어 왔지만 아직까지 정확한 기전에 대한 연구가 미비하여 더 나은 이해와 고찰이 요구된다. 따라서 본 연구에서는 JAK3 저해제가 루게릭병(Amyotrophic lateral sclerosis) 마우스 모델의 생존율을 증가시킨다는 한 연구팀의 연구결과와 성장인자가 풍부한 환경에서 JAK3 억제가 신경전구세포를 신경세포로 분화하는 것을 촉진한다는 본 연구팀의 선행 연구를 바탕으로 성장인자 제거에 의해 유도되는 신경상피세포의 사멸에서 JAK3 억제의 효과를 관찰하였다. 성장인자 제거한 배지에서 배양한 신경상피세포의 세포생존율은 성장인자가 포함된 배양환경에서 자란 세포에 비해 세포 생존율이 감소하였다. 하지만 JAK3 저해제의 처리는 세포 생존율을 증가시켰다. 신경상피세포의 사멸을 trypan blue염색을 통해 관찰하였을 때, 역시 성장인자의 제거는 시간이 지남에 따라 trypan blue에 양성을 보이는 세포(죽은 세포)의 수를 증가시키고 trypan blue에 음성을 보이는 세포(살아있는 세포)의 수를 감소시켰다. 하지만 JAK3 저해제의 처리는 이를 반전시켰다. JAK3 저해제의 효과를 재확인하기 위해서 calcein-AM (살아있는 세포)과 propidium iodide (PI, 죽은 세포)로 염색하여 세포를 관찰했을 때 역시 JAK3 저해제는 calcein-AM에 양성을 보이는 세포의 수를 농도 의존적으로 증가시키는 것을 확인하였다. 하지만 PI에 양성을 보이는 세포의 수는 JAK3 저해제를 처리한 군과 처리하지 않은 군 사이에 차이를 확인할 수 없었다. 또한 살아남은 세포가 어떠한 특성을 지닌 세포인지를 확인하기 위해 세포 표지인자를 이용하여 형광염색법을 사용하였는데 JAK3저해제는 MAP2에 양성을 보이는 신경세포의 생존뿐 아니라 Nestin과 SOX2에 양성을 보이는 신경줄기세포의 생존을 증가시킨 것을 확인할 수 있었다. 그러나 JAK3 저해제는 살아 남아 있는 신경줄기세포의 증식 능력에 영향을 주지는 않았다. 또한 성장인자 제거는 아폽토시스를 유도하는 것으로 알려져 있기 때문에 TUNEL assay를 통하여 아폽토시스가 유도되었는지 확인한 결과, 성장인자 제거에 의해 TUNEL에 양성을 보이는 세포를 관찰할 수 있었고, JAK3 저해제가 TUNEL에 양성을 보이는 세포의 수를 감소시킨 것을 확인하였다. 그리고 아폽토시스 과정에서 중요한 역할을 하는 caspase-3의 활성은 성장인자를 제거한 군에서 증가하는 것을 확인하였지만 JAK3 저해제에 의해 세포생존이 증가한 군에서 caspase-3의 활성이 감소하지는 않았다. 또한 caspase-3 저해제는 신경상피세포의 caspase-3 활성을 유의적으로 감소시키지만 세포 생존율에 영향을 주지 않았다. JAK3 저해제는 성장인자의 제거에 의해 증가한 JAK3의 인산화를 감소시키고 통계적인 의의는 없었지만 세포의 생존 및 사멸의 대표적인 신호경로 중 AKT 활성이 증가시키는 경향과 ERK와 JNK의 활성을 감소시키는 경향을 보였다. 또한 pro-apoptotic bcl-2 family인 bim-EL의 활성은 JAK3 저해제에 의해 감소하는 경향을 보였으며, 신경세포의 접착분자인 N-cadherin과 NCAM의 발현은 JAK3 저해제에 의해 증가되었다. 결론적으로, 성장인자의 제거는 신경상피세포의 아폽토시스를 유도하는데, JAK3 신호의 억제는 이런 아폽토시스로부터 줄기세포의 성격을 지닌 신경상피세포를 보호함으로써 세포 생존을 조절한다. 그리고 이 과정에서 N-cadherin 및 NCAM과 같은 세포 접착분자의 발현을 증가시켜 수상돌기의 성장(neurite-growth)을 동반하는 신경세포로의 분화를 촉진한다. 따라서 JAK3 신호는 신경줄기세포의 생존 및 신경세포의 분화과정에서 중요한 역할을 한다.
