Dichloroacetate, a inhibitor of pyruvate dehydrogenase kinase-1, attenuates hypoxia-induced resistance to 5-fluorouracil in gastric cancer cells

dc.description.abstract | In this study, I investigated whether gastric cancer with hypoxia-induced resistance to 5-fluorouracil (5-FU) could be re-sensitized following treatment with low-dose dichloroacetate (DCA), an inhibitor of the glycolytic pathway. The expression profile of hypoxia-inducible factor-1α (HIF-1α) and pyruvate dehydrogenase kinase-1 (PDK-1) were analyzed in tissues from 10 patients with gastric cancer who had different responses to adjuvant 5-FU treatment. For the in vitro assays, cell viability and apoptosis were evaluated with and without treatment with 20 mM DCA in the AGS and MKN45 cell lines, as well as in PDK1 knockdown cell lines. The expression levels of HIF-1α and PDK-1 were both elevated in the tumor tissues of most patients who showed recurrence after adjuvant 5-FU treatment. Cellular viability tests showed that these cell lines had a lower sensitivity to 5-FU under hypoxic conditions compared to normoxic conditions. Moreover, the addition of 20 mM DCA only increased the sensitivity of these cells to 5-FU under hypoxic conditions, and the resistance to 5-FU under hypoxia was also attenuated in PDK1 knockdown cell lines. In conclusion, DCA treatment was able to re-sensitize gastric cancer cells with hypoxia-induced resistance to 5-FU through the alteration of glucose metabolism and the enhancement of apoptosis. | - |
dc.description.abstract | 본 연구에서는 당분해 경로 억제제인 dichloroacetate (DCA)를 저용량으로 사용하여 저산소영향으로 인한 5-Fluorouracil(5-FU)에 대한 항암제내성을 극복할수 있는 지 여부를 확인 하고자 하였다. 우선 5-FU 보조항암치료요법에서 서로 다른 반응을 보인 위암환자 10 명 의 조직에서 hypoxia-inducible factor-1α (HIF-1α)와 pyruvate dehydrogenase kinase(PDK-1)의 발현 변화를 확인하였고 세포체외 분석에서는 AGS 와 MKN45 그리고 PDK-1 knockdown 세포주에서 DCA 를 처리 하거나 처리 하지 않은 경우에 세포 생존율 과 세포 사멸에 어떠한 영향이 있는 지 확인 하였다. 결과적으로 5-FU 보조항암치료 후 재발 을 보인 대부분의 환자 의 종양 조직에서 HIF - 1α 와 PDK -1 의 발현 수준은 상승 하였다. 그리고 위암세포주에서는 정상 산소조건에 비해 저산소 조건에서 5-FU 에 대해 낮은 민감도를 보이고 있었다. 또한, 20 mM 의 DCA 의 추가는 저산소 조건에서 5-FU 에 대한 위암세포주뿐만 아니라 PDK-1 knock down 세포에서도 항암제감도를 증가시킨다는 것을 확인 할 수 있있다. 결론적으로, 저산소 조건으로 인한 5- FU 에 대한 항암제내성은 DCA 치료제의 포도당대사에 대한 억제와 세포사멸에 대한 강화를 통해 극복 할 수 있다는 것으로 시사되었다. | - |
