Hydrogen peroxide induces cytokinesis failure through DNA damage and Aurora B mislocalization

Abstract
Aneuploidy, an abnormal number of chromosomes that is a hallmark of cancer cells, can arise from tetraploid/ binucleated cells through a failure of cytokinesis. Reactive oxygen species (ROS) have been implicated in various diseases including cancer. However, the nature and role of ROS in cytokinesis progression and related mechanisms has not been clearly elucidated. Here, using time-lapse analysis of asynchronously growing cells and immunocytochemical analyses of synchronized cells, I found that hydrogen peroxide (H2O2) treatment at early mitosis (primarily prometaphase) induced cytokinesis failure significantly. Cytokinesis failure and the resultant formation of binucleated cells containing nucleoplasmic bridges (NPBs) seem to be caused by increases in DNA double-strand breaks (DSBs) and subsequent unresolved chromatin bridges. I further found that H2O2 induced mislocalization of Aurora B during mitosis. All of these effects were attenuated by pretreatment with N-acetyl-L cysteine (NAC) or overexpression of Catalase. Surprisingly, the PARP inhibitor PJ34 also reduced H2O2-induced Aurora B mislocalization and binucleated cell formation. Results of parallel experiments with etoposide, a topoisomerase IIα inhibitor that triggers DNA DSBs, suggested that both DNA DSBs and Aurora B mislocalization contribute to chromatin bridge formation. Aurora B mislocalization also appeared to weaken the "abscission checkpoint". Finally, oncogenic K-RAS stimulated ROS generation in HeLa cells and induced increase in binucleated cell population, which were greatly inhibited by antioxidant NAC or overexpression of Catalase. These results suggested aneuploidy induced by ROS might play an essential role in the oncogenesis at the real situation. In conclusion, I propose that a ROS, mainly H2O2 increases binucleation through unresolved chromatin bridges caused by DNA damage and mislocalization of Aurora B, the latter of which appears to augment the effect of DNA damage on chromatin bridge formation.

하나의 세포당 염색체의 수가 2n이 되지 않는 것을 이수성 (aneuploidy)이라고 하며, 이수성은 암의 기본특성 (hallmark) 중 하나로 알려져 있다. 세포질 분열 (cytokinesis)의 실패로 만들어진 사배체 (tetraploid)/이핵 세포 (binucleated cells)는 이수성 발생의 기전 중 하나로 받아들여지고 있다. 따라서 세포질분열의 이상에 의한 사배체/이핵세포의 생성은 암발생의 한 기전이 될 수 있다. 활성산소 (reactive oxygen species: ROS)는 암을 포함한 다양한 질병들과 관련 되어있다. 하지만, 세포질 분열과정과 관련하여 활성산소가 어떤 역할을 하는지, 그리고 그 기전이 무엇인지는 잘 알려져 있지 않다. 우선 활성산소가 세포질분열에 미치는 영향을 관찰하기 위해 Time Lapse 분석법을 시행한 결과, 과산화수소 (hydrogen peroxide: H2O2) 가 처리된 시점에 세포분열 중 이던 세포에서 유의하게 세포질분열 실패 현상이 증가하는 것을 관찰하였다. 이 결과를 다시 한번 확인하기 위해서 각각 G1기, S기 G2기, M기에 고정(synchronize)시킨 뒤 과산화 수소를 처리하였다. 그 결과 M기, 특히 초기 M기에 위치한 세포 들에서 세포질 분열 실패가 의미 있게 증가 하고 결과적으로 이핵 세포를 형성 한다는 것을 알 수 있었다. 그래서 나는 세포질 분열 실패가 어떻게 일어나는 지를 확인 해 보았고, 과산화수소를 처리하였을 때, DNA double strand breaks (DSBs)와 chromatin bridge의 증가가 세포질 분열을 방해하고, 그 결과로 nucloplasmic bridge (NPB)를 가진 이핵 세포가 형성 된다는 것을 확인 할 수 있었다. 이러한 현상들은 활성산소를 줄여줄 수 있는 N-acetyl-L-cysteine (NAC)이나 과산화 수소를 분해 시키는 Catalase를 과 발현 시켰을 때 감소하였기 때문에, 과산화 수소가 이 현상들을 증가시키고 있다는 것을 입증할 수 있었다. 게다가, M기에 위치한 세포에 과산화 수소를 처리하였을 때, Aurora B의 위치가 비정상적인 것을 확인 하였고, 이 현상 역시 NAC에 의해서 감소하는 것을 확인 하였다. 하지만 Aurora B의 phosphorylation은 감소하지 않았었다. 이러한 Aurora B의 위치 이상만으로 세포질 분열 실패를 유도하는데 기여를 하는지 확인하기 위해 etoposide와 Aurora B의 위치를 비정상적으로 유도 시킬 수 있는 Haspin inhibitor인 CHR-6494를 활용하여 비교 실험하였었다. 결국, DNA DSBs가 있을 때, Aurora B의 위치이상은 chromatin bridge를 형성하는데 기여한다는 것을 알 수 있었고, 과산화수소수를 세포에 처리하게 되면, DNA damage뿐만 아니라, Aurora B의 위치이상을 함께 동반하여 많은 양의 chromatin bridge를 형성하게 되고, 결국 세포질 분열을 방해한다는 것을 알 수 있었다. 게다가 세포질 분열의 마지막 단계에서 chromatin brides가 존재할 경우 세포질 분열 실패에 의한 이핵 세포 형성을 막기 위해서 “abscission checkpoint” 로써 Aurora B의 activity나 정확한 위치 이동이 중요하다고 알려져 있다. 하지만, 이 연구에서 과산화 수소에 의해서 Aurora B의 위치에 이상이 오게 되기 때문에, 아마도 abscission checkpoint가 약해지는 것도 한가지 원인이라고 여겨 진다. 놀라운 점은 과산화 수소와 PARP억제제인 PJ34를 같이 처리하면 Aurora B의 위치이상이 회복되는 것을 확인하였고, 이핵 세포 형성 역시 감소하였었다. 이 관찰의 의미를 확인하기 위한 추가실험이 현재 진행되고 있다. 마지막으로, KRAS가 이핵 세포형성을 증가시키는데에도 역시 과산화수소가 작용하는 것을 볼 수 있었다. 결론적으로, 과산화수소를 M기에 처리하게 되면 DNA damage와 Aurora B의 위치이상을 증가시키면서, chromatin bridge 형성이 증가 되고, 세포질 분열에 방해를 주게 되어, 결국 이핵 세포가 형성 된다는 것을 밝힐 수 있었다. 그리고, 과산화수소에 의해서 증가하는 Aurora B의 위치이상은 DNA damage에 의해서 증가하는 chromatin bridge의 형성을 더 증가 시키는데 도움을 주는 것이라고 여겨진다.
