Abstract
It has been widely accepted that apoptosis is anti-inflammatory because of being removed by immune cells, in contrast, necrosis is pro-inflammatory. However, our previous work has definitely shown that it is possible for pro-inflammatory damageassociated molecular patterns (DAMPs) to be released during caspase-dependent cell death. Extracellular exosomes released by cells are increasing interests as intracellular signaling molecules. For pursuing the inflammatory mechanism of caspase-dependent cell death, therefore, we analyzed the characteristics of extracellular vesicles (EV) released from dying cells and their roles for macrophage activation. Isolated vesicles from caspasedependent cell death are 129.6 +/- 52.2 nm and highly expressed exosome markers such as CD63, LAMP1, and Rab7. But this vesicle is not involved in Endosomal Sorting Complex Required for Transport (ESCRT) pathway because of not containing Alix protein. The exosome-like vesicles highly induced Il1β mRNA and proteins from mouse bone marrowderived macrophage. The release of the vesicles was significantly dependent on caspases because the cells knock-downed with caspase 3, 7, or 9 showed decreased release of vesicles and reduced Il1β induction. Data confirm that Il1β is induced by sphingosine-1-phosphate (S1P) of lysophospholipid and crosstalk between S1P and S1P receptor 3 regulates biogenesis of exosome. These results suggest that apoptotic cell death also can have DAMP-released-immunogenicity like necrosis, which can be mediated by the exosome-like EVs released from the dying cells.

세포고사는 케스페이즈(caspase)라는 단백질 분해효소에 의해 일어나는 일련의 계획적인 자살과정으로 DAMP (Damage-associated molecular pattern)를 분비하지 않고 잔재들은 대식세포 등의 면역세포에 의해 제거가 되기 때문에 염증을 유발하지 않는다고 알려져 왔으나 최근의 연구에서 케스페이즈의 활성과 또 다른 시스테인계열의 단백질 분해효소인 칼파인(calpain)에 의한 DAMP와 뉴클레오좀의 분비를 주요 소견으로 하는 세포사멸기전이 존재함을 규명되었다. 세포 안에서 형성된 endosome이 Intra-luminal vesicle (ILV)를 포함하는 Multi-vesicular body (MVB)로 성숙한 뒤 세포막과 융합하여 밖으로 분비되는 엑소좀은 지질, 단백질, 핵산 등을 포함하고 있어 최근에 세포 간 신호전달물질로 각광받고 있다. 본 연구는 케스페이즈 의존성 세포사멸에서 분비되는 엑소좀의 특징과 엑소좀에 의한 골수 유래 대식세포의 분화를 알아보았다. 먼저 초고속 원심분리 법으로 분리한 vesicle의 크기는 129.6 +/- 52.2 nm로 이전에 보고된 엑소좀의 크기와 일치했고 엑소좀의 marker인 CD63, CD9, LAMP-1를 발현하고 있었다. 따라서 분리한 vesicle은 엑소좀과 유사하였으며, 본 연구자가 분리한 엑소좀은 Alix를 발현하지 않으므로 Endosomal Sorting Complex Required for Transport (ESCRT) complex를 이용해 합성되는 엑소좀과는 다른 합성 경로로 합성됨을 확인하였다. 분리한 엑소좀을 골수 유래 대식세포에 처리 시 IL1b의 전사와 분비를 촉진시키는데 Caspase 3, 7, 9를 knock-down시킨 세포에서 엑소좀의 분비는 줄었고 대식세포에서 IL1b의 분비도 줄었다. 엑소좀내 지질이 IL1b의 유도에 관여함을 확인하였는데 특히 생물학적 활성 대사체인 sphingosine-1-phosphate (S1P)에 의해 IL1b가 유도되었을 가능성을 확인하였고 S1P-S1P receptor signaling axis가 케스페이즈 의존성 관련 엑소좀의 분비에 관여함을 증명하였다.
