NFE2L1 promotes hepatoma cell invasiveness via STX12

Abstract
Mitochondrial dysfunction is an important metabolic feature in human cancer. However, underlying mechanisms how mitochondrial dysfunction affects tumorigenesis remain unclear. To address the role of transcriptomic regulation by mitochondrial defects in liver cancer cells, gene expression profiling for three different cell models of mitochondrial defects was performed previously. By comparing gene expression in the mitochonrial defects models, 10 common mitochondrial defect (CMD)–related genes were identified that may be responsible for retrograde signaling from cancer cell mitochondria to the intracellular transcriptome. Among the CMD genes, NFE2L1 was found that a key regulator to regulate hepatoma invasiveness and was regulated by ROS which is second messenger derived from defective mitochondria. To elucidate how mitochondrial defect regulated NFE2L1 transcription, we selected 11 transcription factors (TFs) that could bind to promoter region of NFE2L1 by using TRANSFAC program. By monitoring NFE2L1 mRNA and protein levels after knockdown of the TFs, I identified that STAT3 regulated NFE2L1 transcription. Interestingly, STAT3 transcriptional activity was higher in SNU354 and SNU423 cells with mitochondrial defects and it was enhanced upon exogenous H2O2 treatment by phosphorylating at Tyr 705 residue. ROS-mediated phosphorylated STAT3(pSTAT3-Y705) regulated NFE2L1 promoter activity and expression. In addition, by performing an NFE2L1-regulated gene expression profiling and association analysis, Syntaxin 12 (STX12) was identified as a key downstream effector of NFE2L1. Also, STX12 was regulated by ROS/STAT3/NFE2L1 axis in hepatoma cells and could promote hepatoma cell invasiveness. Taken together, mitochondrial respiratory defects and subsequent retrograde signaling, particularly the NFE2L1-STX12 pathway, play pivotal roles in liver cancer progression.

미토콘드리아 호흡기능 손상은 암세포의 중요한 대사적 특징이다. 하지만, 미토콘드리아 기능 손상이 암화과정에 어떤 영향을 미치는지에 대해 아직 완전히 설명되지 않았다. 간암세포에서 미토콘드리아 기능 손상에 의한 전사적 조절 기전을 밝히기 위해 미토콘드리아 호흡기능 손상을 유도한 세포모델을 이용하여 유전자 발현 프로파일링을 수행한 것이 보고되었다(Lee et al., 2015). 미토콘드리아 손상에 특이적으로 발현하는 10개의 미토콘드리아 기능 손상 유전자가 선별되었고, 이 중에서도 NFE2L1이 간암세포의 악성화를 조절하는 핵심 유전자이면서, 손상된 미토콘드리아에서 나온 2차적 신호전달자인 ROS에 의해 조절되는 것이 밝혀졌다. 미토콘드리아 손상이 NFE2L1의 전사를 조절하는지 밝히기 위해 TRANSFAC program 을 이용하여 NFE2L1 promoter 부위에 결합할 수 있는 11개의 전사인자를 선별하였다. 전사인자들을 knock-down하여 NFE2L1의 mRNA와 protein의 발현을 확인한 결과, STAT3가 NFE2L1의 전사를 조절하는 상위 전사인자인 것을 확인하였다. 흥미롭게도, 미토콘드리아의 기능이 손상된 SNU354, SNU423 세포에서 STAT3의 전사 활성이 높아있으며, H2O2 를 처리 시 ROS 생성이 증가함에 따라 STAT3의 705번째 잔기인 tyrosine의 인산화가 증대되었다. ROS를 매개로 인산화된 STAT3가 NFE2L1의 전사를 조절하는 것을 확인하였다. 더 나아가, NFE2L1의 조절을 받는 하위 유전자를 찾기 위해 유전자 발현 프로파일링과 상관관계를 분석한 결과, Syntaxin12(STX12)가 NFE2L1의 핵심 하위 유전자로 발굴되었다. 또한, STX12는 간암세포에서 ROS/STAT3/NFE2L1 axis에 따라 조절되고 간암세포의 침윤 활성을 촉진하는 실질적인 작동자인 것을 확인하였다. 결론적으로, 미토콘드리아 호흡기능 손상과 그에 따른 회귀신호체계 중 특히 NFE2L1-STX12 pathway는 간암 세포 침윤 활성에 중추적인 역할을 한다.
