Effectiveness of autogenous tooth bone used as a graft material for regeneration of bone in minipig

Abstract
Purpose: The aim of this study was to evaluate the effect of autogenous tooth bone used as a graft material for regeneration of bone in artificial bony defect of the minipigs.

Material and Methods: Five healthy minipigs, weighing about 35-40 ㎏, were used in this study. Four standardized artificial two-walled bony defect, 5㎜ sized in length and depth were made on bilateral parial edentulous alveolar ridge on mandible of minipig, and autogenous tooh bone was augmented in right side as a experimental group. On the other hand only alloplastic bone graft material HA was grafted with same size and same manner in left side as a control group. All minipigs were sacrificed at 4 weeks after bone graft and histologically evaluated under light microscope with Haematoxylin-eosin staining. Specimens were also semi-quantitatively evaluated via histomorphometric study. Percentage of new bone over total area was evaluated using digital software for area calculation.

Result: All specimens were available but one in the left side (control group) and two in the right side (experimental group) were missing during specimen preparation. As a result of this experiment, bone formation and remodeling process were examined in all experimental group and the control, with more regenerative capacity in the experimental group. Mean percentage area for new bone were 43.74±11.96 % and 30.79±2.93 % for experimental and control groups, respectively.

conclusion: We hereby report that autogenous tooth bone is a good alternative to autogenous bone with expectation of possible clinical feasibility of autogenous tooth bone graft in reconstruction of bony defect.

목적:이 연구는 minipig의 골 결손부에서 자가치아뼈 이식재를 사용한 골이식술시 골형성능 효과에 대해 알아보고자 한다.

재료및 방법:35-40㎏정도의 건강한 다섯 마리의 수컷 미니피그를 이용(24개월)하였으며 하악 양측 소구치및 대구치를 발치하여 자가치아뼈 처리를 하였다.

그후 무치악 하악의 오른쪽에 2개의(각기 5㎜의 높이와 깊이를 가지는)인위적인 bony defect를 형성하여 실험군,반면에 왼쪽은 2개의 대조군으로 하여 실험을 실시하였고 실험군에는 자가치아뼈 이식을,대조군은 Bongros-HA를 이식하였다.미니피그는 이식 4주후 희생시켰으며 결과는 H-E 및 MT염색을 통해

관찰하였다.결과:대조군과 실험군의 골형성 양상을 조직학적으로 비교시 두그룹 모두 신생골형성이 관찰되었다.자가치아뼈 이식재를 넣은 실험군에서 더 활발한 골개조과정을 확인할 수 있었으나 통계적으로 유의한 차이는 보이지 않았다.

결론:위와 같은 결과들을 통해 자가 치아뼈 이식재는 앞으로 더 많은 연구가 필요하겠지만 골결손부에서 골형성을 위한 효과적인 골이식재로서 임상적 유용성이 있을 것이라 사료된다.
