dc.description.abstractObjective: To ascertain the expression of transforming growth factor (TGF)-β receptors in normal human endometrium during the menstrual cycle, and the action of TGF-β and peroxisome proliferator-activated receptor (PPAR)-gamma during endometrial decidualization using cultured human endometrial stromal cells.
Methods: Human endometrial tissues were examined immunohistochemically for the expression of TGF-β receptors and Smad. Western blotting, confocal microscopy and viable cell counting were performed on cultured human endometrial stromal cells which were treated with TGF-β(10ng/mL) and PPAR-gamma agonists (Rosiglitazone(R) 50 nM). Thereafter we compared the effect of TGF-β and PPAR-gamma on the Smad phosphorylation, prolactin expression, and cellular proliferation in vitro human endometrial decidualization.
Results: The results revealed significantly increased expression of both TGF-β receptor-I and -II proteins in the secretory stromal cells compared to the epithelial cells of human endometrium. The degree of expression and translocation into the nucleus of the phosphorylated Smad2/3 was also increased in the secretory endometrium compared to the proliferative endometrium. In the stromal cell culture, the decidualization process associated with TGF-β and pSmad is inhibited by the PPAR-gamma agonist. In contrast to the PPAR-gamma agonist, TGF-β inhibits cellular proliferation.
Conclusion: TGF-β /Smad signaling pathway is essential for endometrial decidualization and closely related to cellular differentiation. PPAR-gamma plays a conflicting role by directly acting on the Smad protein and blocking the TGF-β /Smad signaling pathway.

dc.description.abstract목적: 정상 생리 주기에서 자궁내막에서의 TGF-β수용체의 발현과 배양된 인간의 자궁내막을 이용하여 자궁내막의 탈락막화 과정에서 TGF-β와 PPAR-gamma의 작용을 알아보고자 한다.
연구방법: 인간의 자궁내막조직에서 TGF-β와 Smad 발현을 면역조직화학적 방법을 통해 비교하였다. 자궁내막의 기질세포를 배양하여 TGF-β(10ng/mL)와 PPAR-gamma agonist (Rosiglitazone(R) 50 nM)를 처리하여 Western blotting과 동초점 현미경, 생활성 세포의 수를 관찰하여 자궁내막의 탈락막화 과정에서 Smad, prolactin, 세포증식에 미치는 이들의 작용을 비교하였다.
결과: 인간자궁내막조직에서 분비기에 TGF-β수용체 I, II형의 발현은 상피세포보다 기질세포에서 두 수용체가 모두 높게 발현되었다. 인산화된 Smad2/3의 발현과 세포핵 내로의 이동은 증식기보다 분비기에 더 높게 발현되었다. 기질세포 배양시 TGF-β의 pSmad를 통한 탈락막화 현상은 PPAR-gamma agonist에 의해 상쇄되었으며 PPAR-gamma agonist와 달리 TGF-β는 세포 수의 증식을 억제시켜 세포분화 즉, 탈락막 현상에 관여하는 것을 확인할 수 있었다.
결론: TGF-β는 Smad의 과정을 통해 자궁내막 기질세포의 탈락막화에 관여하고 PPAR-gamma는 TGF-β/Smad 신호전달 경로를 차단함으로써 TGF-β의 작용을 억제한다.
