Study on Association between Single Nucleotide Polymorphisms of MMP7, MMP8, MMP9 Genes and Development of Gastric Cancer and Lymph Node Metastasis

Abstract

목적: MMP family 유전자들은 다양한 인종들에서 암의 발생전이, 침윤과 연관성이 있음이 보고되었다. 이번 연구는 MMP7 rs11568818, MMP8 rs11225395, MMP9 rs17576, rs2250889 다형성과 한국인의 위암 발생, 림프절 전이와의 연관성을 조사하였다.

대상 및 방법: 153명의 위암 환자군과 326명의 만성 위염 환자군으로부터 DNA를 추출하였다. 그 후 MMP7 rs11568818, MMP8 rs11225395, MMP9 rs17576, rs2250889 단일염기다형성을 Golden Gate® 방식으로 유전자형을 분석하였고, PLINK를 이용하여 로지스틱 회귀분석을 하였으며 SAS에서 카이제곱검정을 통해 대립인자와 질병과의 연관성을 분석하였다.

결과: 4개 단일염기성다형성은 만성 위염 환자군과 위암 환자사이에 의미있는 연관성은 없었으며, 만성 위염 환자군과 림프절이 있는 위암 환자군 사이에서도 유의한 차이를 발견할 수 없었다.

결론: 한국인에게서는 전이에 관련된 MMP family 유전자인 MMP7 rs11568818, MMP8 rs11225395, MMP9 rs17576, rs2250889 단일염기다형성은 위암 발생, 림프절 전이와는 연관성이 없을 것으로 생각된다.

Background/Aims: Matrix metallopeptidase (MMP) is known to be involved in tumor invasion and metastasis of cancer. This study investigated the association of MMP7 rs11568818, MMP8 rs11225395, MMP9 rs17576 and rs2250889 with gastric cancer (GC) development and lymph node metastasis (LNM).

Methods: Samples were obtained from 326 chronic gastritis (CG) and 153 GC patients and genotyped by using the GoldenGate® method. Chi-square test was performed to identify the difference of allele distribution between each group (CG vs. GC; CG vs. with LNM GC). The associations of genotype with risk of GC and LNM were estimated by odds ratio and the 95% confidence interval was calculated by logistic regression adjusting for age and sex.

Results: The allele and genotype frequencies of MMP7 rs11568818, MMP8 rs11225395, MMP9 rs17576 and rs2250889 were not associated with the development of GC and LNM.

Conclusions: In summary, MMP7 rs11568818, MMP8 rs11225395 MMP9 rs17576 and rs2250889 were not associated with the GC development and LNM in Korean population.
